The present study was undertaken to examine the long-term effect of continuous light and constant temperature and their interaction on growth, feed intake, gill Na+, K+-ATPase (NKA) activity and early sexual maturation in Atlantic salmon pre- and post-smolts. The fish (mean initial weight = 15.9 g ± 0.4 SE) were reared on two photoperiods (continuous light, LL and simulated natural photoperiod, LDN, 60°25′N) and on two constant temperatures (average 8.3 and 12.7 °C) from June to July the following year. This resulted in four experimental groups abbreviated as LL8, LL12, LDN8 and LDN12. Growth in freshwater was highest in the LL12 group and final weight of this group was 70–330 % higher than in the other experimental groups, and our findings further demonstrate that the growth-enhancing effect of continuous light alone corresponds to a 4.5° increase in temperature. Overall, the highest feed intake was registered in the LL12 group, whereas no differences in feed intake or growth were observed between the LL8 and LDN12 groups, and the lowest feed intake and growth in the LDN8 group. Both temperature groups on LL developed peak levels in gill NKA activity in October–November, 4–5 months prior to the natural season for the parr–smolt transformation. Fish at 12 °C showed peak levels in NKA activity 4–6 weeks before the fish on 8 °C. The proportion of mature males was higher at 12.7 °C (66 %) compared to 8.3 °C (11 %). Highest maturation was seen at LL12 (82 %). For the salmon industry, this means that long-term rearing at LL and 12.7 °C will improve growth but also leads to higher maturation proportion. By rearing fish at LL8, it is, however, possible to achieve high growth and low maturation simultaneously.  相似文献

Two major structural elements of a cell are the cytoskeleton and the lipid membranes. Actin and cholesterol are key components of the cytoskeleton and membranes, respectively, and are involved in a plethora of different cellular processes. This review summarizes and discusses the interaction of alphaherpesviruses with actin and cholesterol during different stages of the replication cycle: virus entry, replication and assembly in the nucleus, and virus egress. Elucidating these interactions not only yields novel insights into the biology of these important pathogens, but may also shed new light on cell biological aspects of actin and cholesterol, and lead to novel avenues in the design of antiviral strategies.  相似文献

Glässer’s disease is a re-emerging swine disease characterized by a severe septicaemia. Vaccination has been widely used to control the disease, although there is a lack of extended cross-protection. Trimeric autotransporters, a family of surface exposed proteins implicated in host-pathogen interactions, are good vaccine candidates. Members of this family have been described in Haemophilus parasuis and designated as virulence-associated trimeric autotransporters (VtaA). In this work, we produced 15 recombinant VtaA passenger domains and looked for the presence of antibodies directed against them in immune sera by immunoblotting. After infection with a subclinical dose of H. parasuis Nagasaki, an IgG mediated antibody response against 6 (VtaA1, 5, 6, 8, 9 and 10) of the 13 VtaA of the Nagasaki strain was detected, indicating that they are expressed in vivo. IgA production against VtaA was detected in only one animal. VtaA were more likely to be late antigens when compared to early (Omp P5 and Omp P6) and late (YaeT) defined antigens. Antibody cross-reaction with two orthologs of Nagasaki’s VtaA5 and 6, VtaA15 and 16 of strain HP1319, was also detected. No antibodies against VtaA were detected in the sera of animals immunized with a bacterin of the Nagasaki strain, suggesting poor expression in the in vitro conditions used. Taken together, these results indicate that VtaA are good candidate immunogens that could be used to improve H. parasuis vaccines. However, their capacity to confer protective immunity needs to be further studied.  相似文献

Aim of the study was to identify changes within xylem structure of grafted trees which could possibly lead to a higher risk of breakage. Therefore, 83 trees from the genera Prunus, Sorbus, Carpinus, Quercus, Robinia and Crateagus were examined at the macro- and microscopic level. All wood samples showed changes in their xylem structure in the areas at or around the contact zone of stock and scion. Cells were deviated in different angles from the stem axis in the tangential direction. Furthermore, whirled grain areas were found adjacent to the contact zone. Abnormal xylem structures could be found in various quantum's. Despite the observed changes in the xylem structure, the risk of breakage is not significantly increased. An e-mail based survey among 496 administrations of rural districts in Germany and bigger towns all over Germany including 64 botanical gardens and arboretum, revealed only nine cases of stem breakage at the contact zone of grafted trees. These breaks were found in the genera Tilia, Fagus and Robinia. Further studies are recommended to gain more knowledge about how xylem structure develops during the lifespan of a tree under stress conditions, especially in urban environments.  相似文献

Stemphylium vesicarium, the causal agent of brown spot of pear, overwinters in the leaf residues of pear and herbaceous plants of the orchard floor. Pseudothecia of the teleomorph, Pleospora allii, are formed on these residues where they produce ascospores. New methods were tested aimed at reducing this overwintering inoculum and increasing the efficacy of control of brown spot of pear. Sanitation methods were evaluated in nine trials in Girona (Spain) and Ferrara (Italy) over a 4-year period. The sanitation methods were leaf litter removal in December to February, and application of biological control agents (commercial formulates of Trichoderma spp.) to the orchard ground cover from February to May. Fungicides were also applied to the trees during the pear-growing season, scheduled according to the BSPcast model. The different methods were tested as stand-alone applications or in combination. All methods consistently reduced the disease incidence at harvest on fruit with an efficacy between 30 to 60% for leaf litter removal and more than 60% for the combination of leaf litter removal and biological control. Efficacy of sanitation alone (leaf litter removal and biological control) in reducing the brown spot level on fruit was similar in most of the trials to the efficacy obtained when fungicides were applied alone. However, integration of sanitation methods and fungicides did not improve the efficacy of disease control over the level provided by fungicides alone.  相似文献

Scientific studies using direct agricultural air quality (AAQ) measurement have experienced revolutionary changes since such a study was first reported in the early 1950s. Comprehensive AAQ research with multi-institutional or international collaboration, long-term monitoring, and multi-pollutant measurement has been conducted with greater frequency. On-site computer system (OSCS), consisting of data acquisition hardware, a personal computer, and custom-developed AAQ research software has become an important tool in these studies. The role of the OSCS has been expanded from exclusively data acquisition to a combination of data acquisition, control, regular and advanced data processing, and communication. This paper studied the general and specific requirements of OSCS in AAQ research and introduced a system with new methodology and technology for high level quality assurance and quality control (QAQC).Data acquisition in AAQ studies is needed for five main categories of instruments and sensors: pollutant concentrations, air exchange, weather conditions, building status, and measurement system status. Regular data processing consists of converting electrical signals to engineering units, averaging data over a given period of time, and performing data correction. Advanced data processing includes calculating location-shared analyzers and sensors (LSAS) data and instrument calibration data and generating daily reports in real-time or upon user requests. The primary control needs in AAQ research centers around the LSAS using multi-point air sampling system. Controls for sampling tubing heating and equipment cooling are often required. Computer communications to monitor system status, deliver alarms and data, and provide remote diagnosis and controls help to reduce operation costs and increase research quality and data completeness.An OSCS that offered flexibility, high capacity, user-friendliness, and high level QAQC was developed for field and laboratory AAQ research. A total of 29 OSCSs have been built and used in various AAQ studies in 13 states in USA for handling a total of more than 3.0 billion data points. The system adapted a set of data processing algorithms as well as some novel features, including All-data Display and Dynamic Run-time Configuration (DDRC), Digital Output DDRC, real-time sampling system monitoring and protection, Global Channel, traceable configuration, and post-measurement data processing. The system also integrated two stand-alone instruments, an Innova multi-gas analyzer and a 7-port Environics gas diluter, which are popular in AAQ research. The strengths, weaknesses, and future development of the system are also discussed.  相似文献

Background  

Complete chloroplast genome sequences provide a valuable source of molecular markers for studies in molecular ecology and evolution of plants. To obtain complete genome sequences, recent studies have made use of the polymerase chain reaction to amplify overlapping fragments from conserved gene loci. However, this approach is time consuming and can be more difficult to implement where gene organisation differs among plants. An alternative approach is to first isolate chloroplasts and then use the capacity of high-throughput sequencing to obtain complete genome sequences. We report our findings from studies of the latter approach, which used a simple chloroplast isolation procedure, multiply-primed rolling circle amplification of chloroplast DNA, Illumina Genome Analyzer II sequencing, and de novo assembly of paired-end sequence reads.  相似文献

In 2001, the European Commission introduced a risk assessment project known as FOCUS (FOrum for the Coordination of pesticide fate models and their USe) for the surface water risk assessment of active substances in the European Union. Even for the national authorisation of plant protection products (PPPs), the vast majority of EU member states still refer to the four runoff and six drainage scenarios selected by the FOCUS Surface Water Workgroup. However, our study, as well as the European Food Safety Authority (EFSA), has stated the need for various improvements. Current developments in pesticide exposure assessment mainly relate to two processes. Firstly, predicted environmental concentrations (PECs) of pesticides are calculated by introducing model input variables such as weather conditions, soil properties and substance fate parameters that have a probabilistic nature. Secondly, spatially distributed PECs for soil–climate scenarios are derived on the basis of an analysis of geodata. Such approaches facilitate the calculation of a spatiotemporal cumulative distribution function (CDF) of PECs for a given area of interest and are subsequently used to determine an exposure concentration endpoint as a given percentile of the CDF. For national PPP authorisation, we propose that, in the future, exposure endpoints should be determined from the overall known statistical PEC population for an area of interest, and derived for soil and climate conditions specific to the particular member state. © 2016 Society of Chemical Industry  相似文献

The aim of this study was to investigate the effect of continuous light at different stages during the production cycle of Atlantic halibut Hippoglossus hippoglossus L. on growth, age at first maturity, endocrine parameters and flesh quality. A group of juvenile halibut [mean (SD), initial weight 191.3 g (44.7)] was reared in indoor tanks under ambient temperature conditions for 38 months until harvesting (mean final weight, 4.6 kg). The entire photoperiod experiment was divided into four phases, where the fish in each phase were exposed to either natural photoperiod (62°33'N) or continuous light (L). Thus, the following five photoperiod combinations were tested: (a) Control group (NNNN), (b) Group 2A (NLNN), (c) Group 2B (NNLN), (d) Group 2C (NNNL) and (e) Production group (LNNN). Exposure to continuous light stimulated growth, and the final mean weights of Groups 2A and 2B were 23% and 11% higher than those of the Control group (NNNN). The final plasma 11-ketotestosteron levels were lower in Groups 2A (2.94 ng mL−1) and 2B (2.46 ng mL−1) compared with the Control (5.29 ng mL−1), Group 2C (5.09 ng mL−1) and the Production group (4.78 ng mL−1) during spring 2007 (age 4 years), indicating higher age at first maturity in Groups 2A and 2B. Photoperiod regime had only a minor, and transient, effect on flesh-quality traits of the fish, whereas a significant seasonal effect was seen with a tendency towards increased gaping, lower pH, lower hardness and lower shear force in July compared with December and March.  相似文献
